  • Design and Development of Communication Skills Training in Diabetes Care for General Practitioners in China

    Abstract: General practitioners(GPs) play an important role in diabetes care in primary care as the "gatekeepers "of population health. The management of diabetes can slow its progression,reduce complications and improve patient outcomes,which requires effective communication and collaboration between patients and their doctors. GPs with good communication skills can help to build long-term care relationships with diabetes patients and help them develop effective self-management skills. This paper summarizes the design and development of diabetes communication skills training for GPs guided by research team with multiple theoretical frameworks,including evidence-based findings from systematic review,experiences and ideas of diabetes patients communicating with GPs based on qualitative studies,prioritization of training content for patient-doctor communication in GPs captured by mixed-methods research,in order to provide solutions for high-quality diabetes management in primary care and inform the design of training programmes for GPs based on evidence and medical education frameworks.

  • Correlation Analysis between Different Free Testosterone Index Levels and Clinical Characteristics of Patients with Polycystic Ovary Syndrome

    Abstract: Background  Polycystic ovary syndrome(PCOS)is a common reproductive and metabolic disorder,mainly characterized by hyperandrogenemia. Free testosterone index (FAI) is not only important in the diagnosis of hyperandrogenemia but also closely associated with clinical parameters,such as glucose and lipid metabolism indexes in PCOS patients. Objective  To investigate the association between different levels of FAI and the clinical characteristics of PCOS patients. Methods  A total of 468 patients with PCOS who were admitted to the gynecology outpatient of the First Affiliated Hospital,Heilongjiang University of Chinese Medicine from October 2019 to June 2021 were included in the retrospective analysis and divided into the low FAI level(LFAI) group(FAI ≤ 2.57,n=156),medium level FAI(MFAL) group(2.576.70,n=156) according to the tertile of FAI levels. The general information,sex hormone levels,glucose and lipid metabolism levels,thyroid hormone and uric acid levels of patients in 3 groups were collected. Pearson correlation analysis was used to analyze the correlation between FAI level and clinical parameters of the patients in the three groups. Results  In the HFAI group,FAI was positively correlated with body mass (r=0.301),BMI(r=0.318),waist circumference(r=0.362),hip circumference(r=0.307),WHR(r=0.280),AND(r=0.407),glucose at 60 minutes(r=0.298),glucose at 120 minutes(r=0.279),glucose at 180 minutes(r=0.281),FINS(r=0.415),insulin at 60 minutes(r=0.320),insulin at 120 minutes(r=0.362),insulin at 180 minutes(r=0.447),homeostatic model assessment of insulin resistance(HOMA-IR)(r=0.446),uric acid(r=0.265)(P<0.05),and negatively correlated with sex hormone-binding globulin(SHBG)(r=-0.486)(P<0.05). Conclusion The levels of FAI are closely related to glucose and lipid metabolism indexes in PCOS patients. Therefore,it is crucial to enhance the screening and attention to clinical parameters in PCOS patients with high FAI levels such as glucose and lipid metabolism indexes.

  • Effect and Mechanism of Tongdu Tiaoshen Electroacupuncture Pretreatment-mediated MiR-124-3p/GSK-3β/Cyp-D Signaling Pathway on Mitochondrial Permeability Transition Pore in Cerebral Cortex of Rats with Cerebral Ischemia- reperfusion Injury

    Abstract: Background  The incidence,mortality and disability rates of ischemic stroke are high,and the reperfusion injury after thrombolytic therapy has a great impact on patients. Acupuncture is a characteristic therapy for the treatment of the disease,but the action mechanism remains unclear. Objective  To observe the effect of Tongdu Tiaoshen electroacupuncture (EA) pretreatment on miR-124-3p/glycogen synthase kinase β(GSK-3β)/cyclin D (Cyp-D)signaling pathway and mitochondrial permeability transition pore of cerebral ischemia-reperfusion injury (CIRI) rats,and explore its possible mechanism of prevention and control of CIRI. Methods  June to August 2022,a total of 100 clean SD rats were randomly divided into the sham operation group,model group,EA group,agonist group and EA + inhibitor group,with 20 rats in each group. For 7 d of intervention before modeling,in the EA group and EA + inhibitor group,"Baihui"(GV 20),"Fengfu"(GV 16)and "Dazhui"(GV 14)were selected to perform electroacupuncture 1 time a day for 7 days. For 24h before modeling,miR-124 agonist and inhibitor (5 nmol) were injected into the lateral ventricles in the EA + agonist group and inhibitor group. Except for the sham operation group,the right cerebral ischemia-reperfusion model of rats was prepared by the modified suture method in the rest groups. The right cerebral cortex of rats was taken,the degree of neurological impairment in each group was observed using mNSS scale and TTC staining,the nerve cell injury was observed by TUNEL staining and transmission electron microscopy. The expressions of GSK-3β,p-GSK-3β,Cyp-D,Cyt-C and Caspase-3 in cerebral cortex of each group were detected by immunofluorescence staining,Western blot and real-time quantitative PCR. Results  Except for the sham operation group,all rest groups of rats were successfully modeled. Compared with the sham operation group,mNSS score and infarct volume of brain tissue were increased,mitochondrial structure was seriously damaged,cell apoptosis index was increased,p-GSK-3β positive expression was reduced,Cyp-D positive expression was enhanced,miR-124-3p,Cyp-D,Cyt-C and Caspase-3 mRNA expressions were increased,p-GSK-3β/ GSK-3β ratio was decreased,relative Cyp-D,Cyt-C,and Caspase-3 protein expressions were increased,and MPTP openness degree was increased in the model group(P<0.05).Compared with the model group,mNSS score and relative infarct volume were decreased,mitochondrial structure destruction was relieved,p-GSK-3β/GSK-3β ratio was increased,and relative expression of Caspase-3 protein was decreased in the electroacupuncture group and inhibitor group;the mRNA expression of GSK-3βwas increased in electroacupuncture + agonist group(P<0.05);apoptosis index was decreased,positive expression of p-GSK-3β was reduced and positive expression of Cyp-D was enhanced respectively,miR-124-3p,Cyp-D,Cyt-C and Caspase-3 mRNA expressions were decreased,Cyp-D and Cyt-C protein expressions were decreased,the degree of MPTP openness was decreased in the electroacupuncture group,inhibitor group and electroacupuncture + agonist group(P<0.05). Compared with the electroacupuncture group,p-GSK-3β positive expression was enhanced,Cyp-D positive expression was reduced,miR-124-3p and Cyp-D,Cyt-C,Caspase-3 mRNA expressions were decreased,GSK-3β mRNA expression was increased,Cyp-D and Cyt-C protein expressions were decreased,and MPTP openness degree was reduced in the agonist group(P<0.05);the apoptosis index was increased,the positive expression of P-GSK-3β was reduced,the positive expression of Cyp-D was enhanced,the expressions of miR-124-3p,Cyt-C and Caspase-3 mRNA were increased,the expressions of Cyp-D and Cyt-C protein were increased,and the openness degree of MPTP was increased in the electroacupuncture + agonist group(P<0.05). Conclusion  Tongdu Tiaoshen electroacupuncture pretreatment can alleviate neurological impairment in cerebral ischemia-reperfusion rats,the mechanism may be related to mediating miR-124-3p/GSK-3β/Cyp-D signaling pathway,inhibiting MPTP openness and thus reducing cell injury. The results of this study further verified the therapeutic effect of Tongdu Tiaoshen electroacupuncture in the treatment of CIRI from the mechanism,providing a new scientific basis for preventive treatment of disease of TCM and promote its clinical application.

  • Constructing Supportive Institutional System to Attract Social Investments to Fund Scientific Research

    Abstract: Innovation always originates from scientific research (basic research and applied research). Investing in scientific research can create new products and new industries, and bring continuous and huge profit. It appeals to a wide range of social investment from market to fund scientific research in recent years. Such economical return, however, is often thwarted by the high risk and uncertainty of scientific research. The main obstacle to the social investments funding scientific research is analyzed in this paper and then some examples are presented as well. It illustrates that it is crucial for public policy to shorten the gap between the science and market in order to make it easier for all kinds of social capital to support innovation. The main challenge for policy makers is to determine the priority of innovation and to reduce the risks, and it is of great essence to construct a supportive system for social investment funding scientific research.

  • Research of S&T and Financing Integration Development from Innovation Chain Perspective

    Abstract: S&T and financing integration contributes to improve innovation efficiency and expand profit margin of financial capital. With the shortening transformation period from basic research into practical productive force, this integration extends to the front of the innovation chain. Dominant position in foundation innovation is becoming the focus of major country competition. From the perspective of innovation chain, this study puts forwards a logical framework of S&T and financing integration. For different innovation stages, the study analyzes the technology innovation goals, industrial development characteristics, and the rules of capital operation. Based on the co-evolution processes of S&T and financing, suggestions of S&T and financing integration development in China are provided.

  • 小事情、大幸福:互动仪式链理论视角下服务仪式对品牌福祉的影响

    Abstract: Nowadays, consumers expect that brands could not only satisfy their functional needs, but also bring emotional and spiritual experiences. Thus, how to garner brand well-being, which means consumers access well-being from using and consuming a brand, has been a challenge for both theory and practice. In fact, whether a brand can create well-being could be shaped by positive interactions between a brand and its consumers. Thus, we postulate that service ritual, which is a prevalence brand-consumer interaction in practice, could create and enhance a brand’s ability to deliver well-being for consumers. Drawing from the interaction ritual chain theory, service rituals refer to a fixed sequence of behaviors that involve symbolic icons and meaningfulness, whereas brands include a series of identifying, integrating, and signaling symbols, indicating the optimal service ritual could form a well-being chain which connects a brand and its consumers. Based on this rationale, the current research draws on interaction ritual chain theory, proposes the new concept of brand well-being, defines and confirms core elements of service rituals, investigates the relationship between service ritual and brand well-being, and further examines the moderating roles of value co-creation orientation of a company and self-brand congruency. In general, the current research offers novel insights on brands and branding theories, well-being and positive psychology literature, and service management theories, while providing implications for companies how to build and manage their brands.

  • 辩证领导行为及其对企业创新能力和绩效的影响: 一项基于中国传统文化的领导行为探究

    Abstract: Although the continuous spread of the pandemic and the rapid development of technology has brought challenges for top managers in China, the environmental uncertainty also offers an opportunity for them to practice and develop leadership skills. To answer what leadership behaviors will help top managers improve organizations’ adaptability and performance in a rapidly changing and complex environment, we developed a new leadership construct, namely, dialectical leadership behavior based on dialectical thinking rooted in the Chinese culture.Dialectical leadership behavior is characterized as a strategic leadership behavior including timely adapting organizational strategy and managerial practices to environmental changes, understanding and balancing between management contradictions, and holistically coordinating different departments and resources in organizations. According to upper echelon theory (Hambrick & Mason, 1984), CEOs’ leadership styles have a profound impact on the strategic choice and performance of the organization. We thus expect CEO dialectical leadership behavior to contribute to firm innovative capability and performance through strategic flexibility in a complex and uncertain business environment.To better understand dialectical leadership, we first developed and validated a scale. We found that dialectical leadership behavior was composed of six dimensions: timely adjusting, individualized mentoring, weighing contradictions, balancing kindness and strictness, promoting coordination, and holistically managing. We further distinguished dialectical leadership behavior from other related leadership constructs with theoretical and empirical efforts. Besides, as top managers of Chinese firms displayed a significantly higher dialectical leadership behavior than those of U.S. companies, the cultural uniqueness of dialectical leadership behavior was justified. Finally, we revealed that CEO dialectical leadership behavior in Chinese companies positively affected firm strategic flexibility and ultimately improved firm innovative capability and performance. We make several contributions. First, scholars constantly explore leadership constructs and practice based on the Western culture. However, the cultural difference between the East and the West matters when it comes to the effectiveness of leadership behaviors. Dialectical leadership behavior provides a new perspective for future exploration of indigenous leadership behaviors and influences by incorporating traditional wisdom of Eastern culture and the reality faced by Chinese enterprises. Second, by developing the measurement of dialectical leadership behavior, we contribute to the future investigation of dialectical leadership behavior and its correlations. Additionally, as we found that CEO dialectical leadership behavior is positively related to firm innovative capability and performance, which are critical for enterprises to survive and thrive during the crisis, we lend confidence in applying wisdom from the Chinese traditional culture to current management practice. Finally, by uncovering the role of firm strategic flexibility in mediating the influence of dialectical leadership behavior on firm innovative capability and performance, we extend understandings about the mechanism through which dialectical leadership behavior matters. We also provide the discussion on the limitation of the study and future research directions.

  • Development of Communication Skills Training in Diabetes Care for General Practitioners in China

    Abstract:

The management of diabetes can slow its progression, reduce complications and improve patient outcomes, which requires effective communication and collaboration between patients and their doctors. Good communication skills in general practice can help to build long-term care relationships with people with diabetes and help them to develop effective self-management skills. This paper summarises the research team's design and development of diabetes communication skills training for GPs guided by a variety of theoretical frameworks, evidence-based findings from a systematic review, experiences and feelings of people with diabetes and GPs based on qualitative studies, and mixed methods research to find the prioritisation of training content in communication skills training. The aim of the paper is to provide solution ideas for high-quality diabetes management in primary care and to inform the design training programmes for GPs based on evidence and medical education frameworks.

  • Research on the accumulation rule of main nutrients and isoflavones and their relationship in Pueraria lobata var. thomsonii

    Abstract:

    In order to determine the optimal harvesting period and the application value of each part of Pueraria lobata var. thomsonii and to explore the correlation between the main nutrients and the isoflavones, Tengxian County P. lobata var. thomsonii was used as the material,the contents of isoflavones in different parts , the contents of main nutrients and isoflavones in different growth periods were determined by test box method, enzyme gravimetric method, Soxhlet extraction method, high performance liquid chromatography and other physiological and biochemical techniques,and the relationship between the accumulation of various nutrients and isoflavones was analyzed to clarify the accumulation rule of main nutrients and isoflavonoids and their relationship in P. lobata var. thomsonii. The results were as follows: (1)At the mature stage, the content of genistein in different parts of P. lobata var. thomsonii was not significantly different, but the content of daidzein in leaves was significantly lower than that in middle vine, and the content of total isoflavones, puerarin and daidzein in roots and vines was significantly higher than that in leaves and roots, and the content of puerarin in roots and vines was more than 1.00%. (2)The accumulation of starch, polysaccharide, crude protein, soluble protein and soluble dietary fiber reached the maximum in November and December, the accumulation of insoluble dietary fiber reached the minimum in December, the accumulation of soluble dietary fiber was the largest in August and December, and the accumulation of total isoflavones and puerarin reached the maximum in August and September. (3)The correlation analysis results showed that the accumulation of isoflavones and puerarin was negatively correlated with the accumulation of starch and polysaccharide; The accumulation of isoflavones and puerarin was positively correlated with the accumulation of insoluble dietary fiber. Therefore, it can be concluded that the root head and vine of pueraria powder contain rich total isoflavones, puerarin, daidzein, which has good value for medicinal development. The best harvest time for medicinal using is from August to September, and the best harvest time for edible using are November and December. The accumulation of starch and polysaccharide of P. lobata var. thomsonii is positively correlated with the accumulation of isoflavones and puerarin, while the accumulation of insoluble dietary fiber is negatively correlated with the accumulation of isoflavones and puerarin.This study clarify the accumulation rule and correlation of the main nutrients and isoflavones in P. lobata var. thomsonii, and provides a reference for the comprehensive development and utilization of P. lobata var. thomsonii and the determination of the harvest time.

    Abstract: Bullens et al. left an open problems in CSI-Fish is to devise a identification protocol that allows for the challenge set to be #1;rather than the small set #1;. This paper proposed a zero-knowledge proof scheme based on supersingular isogeny. This scheme addresses the open problem by taking the challenge C as a isogeny , and reduces the soundness error and the size of public key. This scheme can be turned into non-interactive zero-knowledge proof scheme using the Fiat-Shamir transform. Then signature scheme and group signature scheme based on supersingular isogeny can be implemented under the quantum random oracles model. And this paper analyzes the security and correctness of these schemes.

    Abstract: Taking account of the problems that the standard probability hypothesis density filter is difficult to estimate the target state correctly and has high computational complexity when there are multiple targets move close to each other in clutter environments, this paper puts forward a fast close-spaced target tracking algorithm based on the probability hypothesis density. The proposed algorithm first uses an adaptive gate technology to divide the measurement set originated from the real targets from the sensor measurement set, and then uses the real target measurement set to update the predicted intensity. Finally, a detection-guided close-spaced target reweight approach selectively redistributes the inaccurate component weights within the posterior intensity at each discrete time. Experimental results illustrate that the proposed algorithm not only has target tracking with high efficiency, but also has good stability.

  • 海南龙血树WD40转录因子基因DcWD40-1的克隆和表达分析

    Abstract:海南龙血树是国产血竭的主要基源植物,其血竭主要化学成分为类黄酮化合物。目前对血竭的研究集中在化学成分及药理活性分析,对血竭形成的分子机制并不了解。WD40转录因子在类黄酮的积累中具有重要的调控作用。本研究根据海南龙血树转录组数据,利用RT-PCR技术在海南龙血树中克隆了一个WD40基因DcWD40-1,该基因包含一个1 353 bp的开放阅读框,编码450个氨基酸,蛋白质分子量50.77 kD,理论等电点5.71。生物信息学分析显示,DcWD40-1属于WD40蛋白家族成员,具有5个保守的WD40结构域,和其他植物WD40蛋白同源性高,保守性强。利用Genome Walking方法分离了1 503 bp的DcWD40-1启动子序列,该区域具有典型真核生物启动子结构特征,并含有多个应答激素和胁迫的响应元件。表达分析显示,血竭诱导剂能够诱导DcWD40-1的表达,DcWD40-1的变化与血竭形成及类黄酮积累正相关。此外,DcWD40-1也能对茉莉酸甲酯、细胞分裂素、油菜素内酯和UV-B处理做出积极响应。本结果为进一步研究DcWD40-1在类黄酮生物合成中的潜在功能和作用机制奠定基础。

    Abstract:甾体药物是仅次于抗生素的全球第二大类药物,具有抗炎、抗过敏、调节内分泌等重要功效。对甾体母核特定位点进行有效的氧化修饰,是引入药效活性的关键。研究表明,P450酶是催化甾体特异性氧化的一类关键酶家族。目前,电子传递效率和催化特异性是限制P450酶催化功能的重要因素,会导致目标甾体产物产量低、副产物积累严重等问题。因此,本文将围绕提高P450酶的催化效率及特异性方面的研究工作,系统综述工程化P450催化甾体类物质的方法策略和相关研究进展,并对催化甾体化合物的P450酶的设计和优化的未来发展进行展望,为该方面工作的深入研究提供指导。

  • IL-27及其受体对肺成纤维细胞增殖及转化的影响

    Abstract: Objective To investigate the effects of interleukin-27 (IL-27) and its receptor (WSX-1) on the proliferation, transformation and collagen synthesis of the mouse lung fibroblasts. Methods Cultured mouse lung fibroblasts were treated with TGF-β1, recombinant murine IL-27, a IL-27 receptor (IL-27R) overexpression vector IL-27R/pCDNA3.1, IL-27 and IL-27R, or all the 3 combined. MTT assay was used to assess the proliferation of the cells, and RT-PCR and Western blotting were employed to examine the mRNA and protein expressions of a-smooth muscle actin (α-SMA) and types I and III collagen; immunofluorescence assay was used to test the expression and location of α-SMA. Results TGF-β1 promoted the cell proliferation and obviously enhanced α-SMA expression and types I and III collagen synthesis in the fibroblasts. Both IL-27 and IL-27R significantly inhibited the proliferation of the pulmonary fibroblasts and obviously decreased their α-SMA expression and types I and III collagen synthesis, but when combined,they produced no obvious inhibitory effect on TGF-1-induced proliferation and transformation of pulmonary fibroblasts. Conclusion Both IL-27 and IL-27R alone can suppress the proliferation, transformation, and collagen synthesis of mouse pulmonary fibroblasts, but their combined treatment produces no such inhibitory effect because of the neutralization of exogenous IL-27 by IL-27R to result in the failure of activating the cell signaling pathways.

    Abstract: To protect the priority right of the authors and to promote the excellent scientific research articles first preserved and serve for other researchers in our country, Scientific Article Preprint Platform of Chinese Academy Science (ChinaXiv) is proposed to build. It aims to encourage researchers to share their articles on public preprints platform in the first time,and receive peer reviews and suggestions for a wider range of academic communication. Based on the practice and strategic plan of ChinaXiv construction, this paper describes how to build a preprint platform following the international standard operation regulation, including construction background, system construction and service, domain development and cultivation, organization and management mechanism, propagandizing and popularizing mode, development status and future. This paper can provide references for other preprint repositories.

    Abstract:【目的】通过采用语义识别、知识关系计算等方法提升科技文献检索系统的服务功能和效果, 使之能够呈现更加丰富的知识化语义信息, 将更多的知识点和知识关系展现给用户。【方法】应用数据挖掘和关系计算工具,深度识别和抽取科技文献中的语义知识, 分析、计算、构建语义关系, 并将得到的语义知识和语义关系建立多维语义索引树, 设计新的数据组织呈现模型。【结果】研发语义丰富化检索示范系统, 在科技文献检索应用过程中充分揭示语义信息, 丰富检索体验。【局限】选取的试验数据集合不够充足, 缺少其他领域应用对比。【结论】本文模型设计给用户带来更多的知识层面的关联、揭示和导航, 提升了检索系统体验。同时分析了设计模型的不足之处, 探索改进方法。

    Abstract:【目的】拓展以物种为中心的植物物种多样性抽取框架, 探索实现语义知识抽取方法。【方法】结合当前生物多样性抽取的主流研究, 以物种为中心, 设计包含多种实体及实体间关系的知识抽取框架, 利用已有的众多专业数据库, 设计并实现相应的识别方法。【结果】设计以物种为核心的知识抽取框架, 探索实现多种实体及实体间关系的语义知识抽取方法, 拓展植物物种多样性领域抽取内容和思路。【局限】实体识别的完整性和准确性受底层知识库影响较大, 且实体间关系的类型局限于共现、上下位类、语法关系几类, 还需进一步研究。【结论】本研究拓展了植物物种多样性抽取内容和思路, 可有效支持语义检索、科学计算。
