  • Cloning, structural and function analysis of MiMYB44L gene in kernels of Macadamia integrifolia

    Abstract: Macadamia nut (Macadamia integrifolia) is an evergreen nut tree with high economic value. Its kernel is rich in nutrients such as fatty acid and protein, etc. In order to further explore the main regulatory genes related to nutrient formation in M. integrifolia kernels, transcriptomics, gene cloning, fluorescence quantification PCR and bioinformatics techniques were used to screen potential regulatory genes from the kernel transcriptomes of ‘Guire No. 1’ and ‘A4’, which have significantly different nutrient content in M. integrifolia kernels. The results were as follows: (1) Transcriptome analysis showed that 1 667 genes were up-regulated and 1 798 genes down-regulated in ‘Guire No. 1’ kernel compared with those of ‘A4’ kernel and KEGG enrichment analysis showed that the differential genes were mainly in starch and glucose metabolism, amino acid biosynthesis and carbon metabolism. (2) A significant differential gene-LOC122077931 encoding the R2R3-MYB transcription factor MYB44L was discovered. The MiMYB44L gene was cloned in kernels of M. integrifolia variety ‘Guire No. 1’ using RACE technology, which was 1 165 bp in length, 999 bp in ORF in length, and encoded 332 amino acids. (3) Bioinformatics analysis confirmed the presence of the SANT domain in the MiMYB44L protein, a hallmark feature of the R2R3-MYB family. The protein lacked both a signal peptide and a transmembrane domain but featured phosphorylation sites. (4) The protein content in kernels of 10 M. integrifolia varieties was determined. And it was found that the expression of MiMYB44L gene in M. integrifolia varieties with high protein content was significantly higher than that in varieties with low protein content, and the overall correlation coefficient was 0.54, reaching a very significant level. The results of this study provide theoretical guidance for in-depth analysis of the regulatory mechanism of MiMYB44L gene in the formation of protein content in M. integrifolia.

  • 基于改进方法的承德地表水环境质量评价

    Abstract: To objectively reflect the surface water quality status, a combination of the Analytic Hierarchy Process Entropy Weight Method and the weighted average comprehensive evaluation principle were used to improve the fuzzy comprehensive evaluation method. The method was compared with the single factor evaluation, Nemero index, and traditional undefined extensive evaluation methods to evaluate the surface water environmental quality in the Chengde area in 2021. The results showed a significant spatiotemporal difference in the surface water quality. Except for some sections with poor water quality from July to September, the water quality in other sections was in Classes I–III; The main factors affecting surface water quality in the Chengde area were CODMn and TP; The single factor evaluation and the Nemero index methods could not reflect the overall water quality. Compared to the traditional fuzzy comprehensive evaluation method, the improved fuzzy comprehensive evaluation method considers the interaction between various pollution factors and weakened the impact of individual water quality indicators on the evaluation results. It can also rank the category of water quality, making it more suitable for evaluating the environmental quality of surface water in the Chengde area.

  • Comparative Analysis of China's Big Data Policies from the Perspective of Text Similarity

    Abstract: [Purpose/significance] The formulation and implementation of big data policies is an important means for the country to promote the development of the big data industry. Therefore, research on big data policies has received widespread attention from the society.[Method/process] From the perspective of text similarity, the article compares the Big Data Development Action Plan issued by the State Council and the texts of big data policies released in 22 regions.[Result/conclusion] Data shows:the policies formulated by Guangdong Province and Fujian Province are the most complete and comprehensive;open data sharing and security guarantees the highest overall attention in the formulation of big data policies in various regions, showing similarity;regional characteristics are more prominent, showing differences. With the successive release of artificial intelligence policies in various regions, future research on big data policies under the vision of artificial intelligence will become a new direction.

  • Research on Algorithm Risk and Regulation in Intelligent Intelligence Analysis

    Abstract: [Purpose/significance] In recent years, artificial intelligence has brought changes in thinking, concepts, methods and techniques to national intelligence work, making intelligent intelligence analysis gradually become one of the important tasks for the innovation and development of national intelligence. This paper studies the algorithm risk and its regulation in intelligent intelligence analysis to avoid the security risks caused by artificial intelligence algorithms and reduce the factors that restrict the development of intelligence work.[Method/process] Based on the interpretation of the core algorithms and research issues of intelligent intelligence analysis, combined with actual application scenarios, the causes and consequences of algorithmic risk and the interaction between the factors of algorithmic risk are analyzed. Establish a gradual regulatory framework for intelligent intelligence analysis algorithms through pre-assessment, mid-event supervision, and post-event accountability.[Result/conclusion] This article analyzes the formation mechanism of the "intelligence" cocoon room, and proposes a virtuous cycle of algorithm regulation and coordinated development before, during, and after the risks of algorithm black boxes, algorithm defects, and algorithm manipulation in intelligent intelligence analysis, and believes that Facing the two sides of the algorithm is also an effective way to prevent and defuse the risk of the algorithm.

  • Prediction of Critical Pitting Temperature of 316L Stainless Steel in Gas Field Environments by Artificial Neutral Network

    Abstract: 316L stainless steel is widely used for enhancing the pitting resistance of pipelines in gas field. The corrosion environment is complex and diversified in different working districts of gas field. Therefore, it is necessary to develop a model for predicting the pitting resistance of pipelines serving in different corrosive environments. Critical pitting temperature (CPT) is considered as a criterion for evaluating the pitting resistance of stainless steel. Based on a survey on the operation situations of gas field, the relevant data of CPT for 316L stainless steel is acquired by potentiodynamic polarization method in solutions with various Cl- concentrations and pH values, which are selected to correspond with the real environments in operation. Then, an artificial neutral network (ANN) model is developed to predict the CPT, and therewith to compare with the measured data. The results show that the CPT decreases with the increase of Cl- concentration, but on which pH value has little influence. The developed ANN model has good ability to predict the CPT of 316L stainless steel, and can be used for the prediction of CPT in complex environments in gas field. It is also revealed that there is no interactive effect between Cl- concentration and pH value, and Cl- concentration was the main influencing factor on the CPT. Therefore, Cl- concentration will be peculiarly concerned with for the implementation of a corrosion control project in gas field.

  • In Situ Study of Hydrochloric Acid Dew Point Corrosion of Stainless Steel

    Abstract: Hydrochloric dew point corrosion (DPC) behavior of 304 and 316L stainless steels was investigated using a novel DPC simulation set-up with in situ measurements of electrochemical impedance spectroscopy and electrochemical noise. The results indicated that the DPC resistance of 316L stainless steel was superior to 304. The difference of the corrosion performance for the two stainless steels can be ascribed to the following two aspects: the ratio of Cr/(Cr+Fe) is higher and the concentration of Fe is lower in the passive film of 316L stainless steel. There exists Mo in the passive film, which can enhance its pitting resistance of 316L stainless steel.

  • Effect of Pre-treatment on Corrosion Resistance of Titanatezirconate Conversion Coating on Magnesium Alloy

    Abstract: Effect of pre-treatment processes including acid pickling and acid pickling plus alkaline cleaning on the corrosion resistance of achromium-free zirconate-titanate based conversion coating on AZ91D magnesium alloy was systematically investigated. The results indicated that the acid pickling could induce the preferential dissolution of α phase and thereby increase the surface roughness of AZ91D magnesium alloy, but which was not conductive to the enhancement of the corrosion resistance of the conversion coating. However, the acid pickling plus alkaline cleaning could appropriately modulate the surface chemical state of AZ91D magnesium alloy, and therefore effectively improve the corrosion resistance of the zirconate-titanate based conversion coating.

  • Corrosion Failure Analysis of a Tee in a Natural Gas Field

    Abstract: Incident involving failure occurred for a 40 months old upright type tee of gas filed gathering and transmission pipeline in a west natural gas field. The tee consisted of a main pipe of 16Mn steel and branch pipes of bimetal composite of 316L stainless steel+L416 steel. Examinations were performed to identify the cause of the failure, which include visual physical inspection, pipe thickness measurement and acquiring the galvanic corrosion behavior of 16Mn steel and 316L stainless steel in an oxygen-free medium containing chloride ion concentration 150 g/L with CO2 partial pressure 0.1 MPa at 65 ℃ using electrochemical methods, as well as Computational Fluid Dynamics (CFD) analysis of the tee with assumed angles as 90°, 45°and 30°respectively between the branch pipe and main pipe along the direction of inlet using the FLUENT software. Investigations revealed that for the present tee design, there existed galvanic couple of the metallic materials and the straight cutting structure of the tee caused the fluid changes radically. Gas field water gradually adhered to the inner side of the pipe due to the low velocity that caused by the vortex which was formed nearby the pipe explosion site. Corrosion then ensued as CO2 dissolved in the water. The area around the explosion site suffered from large wall shear stress caused by the fluid scouring action. Because of the synergy of galvanic corrosion and fluid flow erosion the pipe gradually thinned, and then perforated.

  • Dew Point Corrosion Behavior of Rolling Q235 Steel Using In Situ Electrochemical Measurement

    Abstract: Dew point corrosion (DPC) behavior of rolling Q235 steel was investigated using simulative equipment and electrochemical noise. The results indicated that mass loss of Q235 steel along the rolling direction (RD) is more than that along the normal direction (ND). The main reason was that the cementite was not so easily broken off along normal direction during the corrosion process because of the net-like characteristic of ferrite and pearlite. And weight loss resulting from galvanic corrosion in the two directions was nearly the same with each other.

  • 多源数据融合视角下的大学生“消费-学业-社交”画像构建研究

    Abstract: [Purpose/Significance] Mining college student data and constructing studnet profiles is conducive to in-depth understanding of students' needs, improving management level, and promoting intelligent service. [Method/Process] Based on the multi-source data mainly generated by the management and service process of colleges and universities, student profiles were developed by focusing on consumption, academic and social indicators, analyzing the characteristics of students, using the Scikit-Learn tool of Python, and applying the K-means clustering algorithms. Empirical research was carried out and representativeness of student portraits from individual and group perspectives was studied. [Results/Conclusions] First, this paper attempts to utilize a new data fusion perspective, by fusing explicit data with implicit data, and generating three-dimensional indicators of consumption behavior, academic behavior, and social behavior. Secondly, in order to solve the problem of single application scenario in previous research, the method of user profile construction is used to realize the fusion of multiple scenarios. Finally, based on the real student data, this study uses K-means clustering algorithm to select groups of students with different characteristics on the basis of previous research. The data of college students is analyzed, and further empirical research is carried out to describe the "consumption-academic-social" profiles of college students. Constructing student profiles from the perspective of multi-source data fusion can effectively provide a basis for decision-making by different units in colleges and universities, such as academic affairs,. Especially in the post-epidemic era, the profiles of college students can detect potential risks in time. The study found that at the individual level, by interpreting the label information of students' portraits, it is possible to understand the 3 aspects of students' consumption, academics and social interaction, and realize dynamic monitoring of individual students. At the group level, through cluster analysis, students with different characteristics can be selected, especially in terms of consumption behavior, and the characteristics of students' activity and stability can be deeply analyzed, which can not only provide a basis for the macro-level observation of students, but also provide new ideas for exploring the correlation between different behavioral elements of students. At the application level, the integration of multi-scenario student profiles can simultaneously realize abnormal identification and early warning, group attention and guidance, and resource planning and adjustment, which greatly broadens the application scenarios of research and improves the energy efficiency of education and teaching management in colleges and universities. However, due to the limitations of data and algorithms, the accuracy and ease of use of student portraits still need to be improved. There are both constraints from practical conditions and insufficient research methods. In future research, more extensive research should be used to improve college student profile construction system, and constantly develop more suitable techniques.

    Abstract: In more than 30 years of practice of poverty alleviation through science and technology, the Chinese Academy of Sciences (CAS) has always been adhering to the concept of “poverty alleviation through science and technology to enhance the selfhematopoietic function of poverty-stricken areas”. Two main approaches, i.e., “science and technology innovation promotes poverty alleviation” and “science and technology think tank serves macro decision-making”, are focused, poverty alleviation is combined with aspiring ambitions and fostering intelligences, and the combination of theoretical research, technological research and development, and decision-making services is insisted. Having been giving full play of the CAS’s advantages of disciplines and platforms, achievements and talents, the poverty alleviation in designated areas has been promoted, and the national poverty alleviation assessment has been supported. Such practices have also actively explored in helping the world in poverty reduction and development, and contributed greatly to the fight against poverty.

  • Influence of Hydrogen Absorption and Desorption on Magnetocaloric Effect of La0.6Pr0.4Fe11.4Si1.6B0.2 Alloy

    Abstract: The La0.6Pr0.4Fe11.4Si1.6B0.2 alloy was produced by using industrial grade raw materials. The phase constituents and thermal magnetic properties of the alloy were characterized by X-ray diffraction with Cu kα radiation and vibration sample magnetism respectively. The influence of hydrogen absorption pressure on magnetocaloric effect and hydrogen desorption process were investigated. Results show that the annealing treated alloy was subjected to hydrogenation at 0.13 and 0.2 MPa, correspondingly its Curie temperature increased to 320 and 321 K respectively. The taking place of dehydrogenation was much easier for the alloy hydrogenated at lower absorption pressure rather than that at higher pressure. The Curie temperature of the alloy hydrogenated at 0.13 MPa and then dehydrogenated at 473K could lower to room temperature range, correpondingly the maximal magnetic entropy of the hydrides decreased while the relative cooling power were increased.

  • 光枝勾儿茶内生真菌多样性及抑菌活性研究

    Abstract: To explore the fungal taxa, distribution and antibacterial activity of Berchemia polyphylla var. leioclada, endophytic fungi of B. polyphylla var. leioclada from Guiyang and Qianxi of Guizhou Province were isolated by tissue fragment separation. Based on the methods of molecular biology and statistics, the endophytic fungi were identificated and their diversity were analyzed. The strains with antibacterial activity were screened by microdilution of broth. The results were as follows: (1) There were 191 fungal strains were isolated, which classified into 3 phyla, 5 classes, 10 orders, 15 families and 19 genera. Among all strains, Phyllosticta、Diaporthe、 Botryosphaeria and Colletotrichum were the dominant genus. (2) The fungal endophytes’ Shannon-Weiner diversity index of Berchemia polyphylla var. leioclada in Qianxi (H'Q=2.112) was higher than Guiyang (H'G=1.801), the Sorenson’s similarity coefficients CsG-Q was 0.923. The Shannon-Weiner diversity index of different tissues was stem (H'S =2.004)> root(H'R=1.764)> leaf(H'L=1.654)>fruit(H'F=1.473). The Simpson was stem(DS=0.826)>fruit(DF=0.813)> root(DS=0.765)>leaf(DL=0.721), the similarity index between stem and leaf was the highest which was CsS-L =0.667. (3) 21 endophytic fungi’ secondary metabolites have inhibitory effect on Escherichia coli, Staphylococcus aureus and Salmonella enterica. The isolate Diaporthe sp. QX4G6 fermentation broth extract have relatively high inhibitory effect, which the minimum inhibitory concentration against Escherichia coli, Staphylococcus aureus and Salmonella enterica were 12.5, 6.25 and 12.5 mg·mL-1, and the minimum bactericidal concentration were 12.5, 6.25 and 12.5 mg·mL-1, respectively. The study revealed rich endophytic fungi resources of Berchemia polyphylla var. leioclada and most of fungi have good antibacterial activity, which lay the foundation for the research and development of natural antibacterial drugs or drug sources for endophytic fungi from B. polyphylla var. leioclada.

  • Effect of microbiota-targeted therapy on intestinal microbiota in patients with irritable bowel syndrome

    Abstract: Background The role of gut microbiota in the pathogenesis and treatment of irritable bowel syndrome (IBS) has attracted attention at home and abroad, and a variety of therapeutic methods targeting the microbiota have been used in clinic. Objective To analyze the effect of therapy targeting microbiota on gut microbiota in patients with IBS, and to provide reference for optimizing the intervention strategy. Methods The web of science core collection, PubMed, EMBASE and Cochrane databases were searched from the establishment of the database to May 20, 2022. The literatures on the changes of gut microbiota after treatment with a single kind of probiotics, prebiotics, antibiotics and fecal microbiota transplantation (FMT) were screened and reported. The data were extracted, and the research results were summarized and reported. Results A total of 3044 articles were retrieved, and 41 articles were included after screening by inclusion and exclusion criteria, including 11 studies on probiotics, 8 studies on prebiotics, 5 studies on antibiotics and 17 studies on FMT. Randomized controlled trials, Rome III, diarrhea predominant IBS and 16S rRNA amplicon sequencing were the most commonly used trial types, diagnostic criteria, IBS subtypes and mcirobiota detection methods. Lactobacillus is the most used probiotic species, which can maintain its activity in the feces of IBS patients. Prebiotics can promote the colonization of probiotics (mainly Bifidobacteria) in feces and colonic mucosa. Rifaximin, an antibiotic acting locally in the intestine, could reduce the species richness of the microbiota, but had no significant effect on the microbial structure. After FMT, the microbial structure of IBS patients tended to move towards that of donors, which can increase the probiotics and reduce the abundance of pathogenic bacteria. Conclusion Treatment targeting microbiota is helpful for the improvement and reconstruction of intestinal microenviroment in IBS patients. In the future, more accurate changes of microbiota after these treatment methods can be further obtained by unifying diagnostic criteria, using more advanced detection technology, and prolonging the follow-up sampling time, so as to provide a basis for clinical development of accurate microbiota-targeted intervention strategies.

    Abstract:利用发射到平流层的球载太阳望远镜来观测太阳磁场演化和监视太阳活动有着得天独厚的优势。首先,在平流层中,球载太阳望远镜对太阳的观测不受来自地球对流层大气中天气现象的干扰,处于无视宁度影响的环境中。这为获取高质量的太阳图像提供了优越的条件。其次,平流层的空气已经十分稀薄,对紫外线的吸收也大为减弱。球载太阳望远镜能够在近紫外波段观测太阳活动和爆发。第三,球载太阳望远镜可以通过回收、升级和再利用来降低其使用成本并提高望远镜的利用率,远比空间观测经济实惠。利用球载太阳望远镜开展对太阳的观测研究在欧美已经有半个多世纪的历史。本文简要地回顾了太阳观测的球载任务发展历史,包括在这期间积累的非常丰富的仪器研制和观测经验,详细地介绍欧洲“日出”(SUNRISE)球载任务的仪器搭载、大量的高分辨观测数据和一系列在此基础上完成的高质量的科研成果,为我国球载望远镜的研制提供重要参考。

  • 基于ARM架构的嵌入式系统在自动气象监测系统的应用:以NVST气象站为例

    Abstract: 自动气象监测系统是现代天文观测台站必备的辅助系统之一,传统自动气象系统是基于微控制器或PC开发的。微控制器多用于工业控制,无法满足多任务和批量数据的快速处理;PC由于高功耗、高成本及低可移动性无法在野外如选择台址时使用。为了克服上述两种开发方式的缺点,本文采用基于ARM架构的嵌入式系统为抚仙湖NVST开发一套低功耗、低成本、高稳定性的自动气象监测系统。本文介绍了NVST气象站的整体设计,软硬件功能以及调试中遇到的问题及解决方法。

  • 射电干扰中基于测距的定位算法研究

    Abstract:对于射电望远镜观测的天文研究,即使很微弱的射频干扰也会导致观测失败或降效。当前射电望远镜台址射频干扰越来越多,需尽快尽量将射频干扰源找到并予以消减。无线定位技术成为查找干扰源时,获取射频发射源准确位置的重要技术手段。基于测距的无线定位算法因其定位精度高,稳定性强而备受关注。本文对几种基于测距的无线定位算法进行研究,分析比对这几种测距定位算法的特点。通过比对,TDOA(Time Difference of Arrival,基于到达时间差)定位算法计算复杂度低,定位精度高;RSSI(Received Signal Strength Indication,基于接收信号强度)定位算法简单易于实现。在此基础上提出了一种基于TDOA定位算法和RSSI定位算法数据融合的测距无线定位算法,该算法在通过TDOA定位算法获得多个初始位置估计值的基础上引入以信号接收强度作为初始位置估计值的权值的数据融合方式,通过线性加权归一化处理获得一个定位精度较高的位置估计值。因信号接收强度易于获得,该数据融合算法在TDOA定位算法基础上增加的计算量较小,且仿真结果表明该算法较单一的TDOA定位算法降低了约20%的定位误差,有效提高了定位精度,便于快速低成本发现射频干扰源。

    Abstract:本试验旨在研究饲粮添加共轭亚油酸(CLA)对开产蛋鸡生产性能、蛋品质、蛋黄硬度和蛋黄脂肪酸组成的影响。采用单因素随机试验设计,选用产蛋率、体重相近的18周龄海兰褐蛋鸡630只,随机分为7组,每组6个重复,每个重复15只鸡,分别饲喂添加0、1%、2%、3%、4%、5%和6% CLA的试验饲粮,预试期1周,正试期8周。结果表明:1)饲粮CLA添加水平≤2%时,对蛋鸡的生产性能、蛋品质无显著影响(P>0.05)。与对照组相比,饲粮添加2%~4% CLA可增加蛋壳厚度(P>0.05);饲粮CLA添加水平≥3%时,产蛋率显著降低(P<0.05),蛋黄硬度显著增加(P<0.05);饲粮CLA添加水平≥4%时,平均日采食量和腹脂率显著降低(P<0.05);饲粮CLA添加水平≥5%时,料蛋比显著升高(P<0.05),蛋黄比例显著降低(P<0.05)。2)1%、2% CLA添加组蛋黄中C16:0、C18:0和饱和脂肪酸(SFA)含量显著低于其他组(P<0.05)。与对照组相比,饲粮CLA添加水平≥4%时,蛋黄中C16:0、C18:0和SFA含量显著增加(P<0.05),C16:1、C18:1、单不饱和脂肪酸(MUFA)和多不饱和脂肪酸(PUFA)含量显著降低(P<0.05)。蛋黄中CLA含量随饲粮CLA添加水平的增加呈显著二次升高(P<0.01),c9,t11-CLA的富集量是t10,c12-CLA的1.47倍。综上,饲粮添加2% CLA,可以改善蛋壳质量,降低蛋黄中SFA含量,蛋黄中可富集一定量的CLA,且不影响蛋鸡的生产性能和蛋品质。

    Abstract:本试验旨在探讨饲粮添加亮氨酸和谷氨酸对肥育猪肌肉脂肪酸组成和脂质代谢相关基因表达的影响。选取平均体重为77 kg左右的三元杂交肥育猪60头,随机分为5个组,每组12头,公、母各占1/2。对照组饲喂基础饲粮,试验组分别在基础饲粮中添加2.05% L-丙氨酸(等氮对照组)、1.0%亮氨酸+1.37% L-丙氨酸(亮氨酸组)、1.0%谷氨酸+1.44% L-丙氨酸(谷氨酸组)、1.0%亮氨酸+1.0%谷氨酸(亮氨酸+谷氨酸组)。饲喂60 d后屠宰采集肌肉样品,检测其中脂肪酸含量以及脂质代谢相关基因mRNA的相对表达量。结果表明:与对照组相比,在背最长肌中,亮氨酸组C18:2n-6和C20:1含量显著增加(P<0.05),C18:0含量显著降低(P<0.05),谷氨酸组C14:0和C16:0含量显著降低(P<0.05),C17:0和C18:2n-6含量显著增加(P<0.05);在股二头肌中,亮氨酸组C16:0含量显著降低(P<0.05),谷氨酸组C18:2n-6含量显著增加(P<0.05)。与对照组相比,亮氨酸组背最长肌中脂肪酸转运蛋白1(FATP-1)mRNA的相对表达量显著上调(P<0.05),亮氨酸+谷氨酸组背最长肌和股二头肌中脂肪酸转运蛋白4(FATP-4)mRNA的相对表达量表达显著下调(P<0.05)。与等氮对照组相比,谷氨酸组和亮氨酸+谷氨酸组背最长肌中脂蛋白脂酶(LPL)mRNA的相对表达量显著上调(P<0.05)。上述结果提示,饲粮添加1.00%亮氨酸或1.00%谷氨酸可调控肥育猪肌肉中脂肪酸组成及脂质代谢相关基因的表达。
