  • Enlightenment and Construction Mode of German Research Data Infrastructure

    Abstract: [Purpose/significance] The research data infrastructure is an indispensable supporting infrastructure for the efficient management, utilization, and efficient operation of digital scientific research. [Method/process] This paper took the German research data infrastructure projects as the research object, and analyzed the construction mode from the perspectives of national and regional task content, construction organisation and related management mechanisms. [Result/conclusion] The study found that the German research data infrastructure has different emphasis at different levels. At the national level, data standards and tools are the main focus, while at the local level, services and training are the main focus. It adopts a strategy of extensive cooperation and complementary capabilities in construction. And in management, it has the characteristics of focusing on unified management and supervision. This paper concludes with suggestions for China's research data infrastructure construction.

  • Current Situation and Improvement Suggestions of Archives Management of CAS Field Stations

    Abstract: The archives resources of field stations of Chinese Academy of Sciences (CAS) are important basic scientific and technological resources of the country. In the new era, China has put forward new requirements for the work of scientific research archives. The open sharing and network construction of field stations pose new challenges to the archives management of field stations. In order to deal with the new problems of field station archives management, based on the investigation of the data, documents, and archives management of 137 field stations and 50 supporting affiliations of CAS, this paper analyzes the current situation and problems of field station archives management, and explores the suggestions for improvement. This paper has certain practical and guiding significance for improving the archives management level of CAS field stations, enriching the content of scientific research archives, and building a basic science and technology resource system.

  • Thoughts on Future Development of Field Observation and Research Network of Chinese Academy of Sciences

    Abstract: Acting as a distributed field science and technology infrastructure, the field station network of Chinese Academy of Sciences (CAS) has evolved to be an important scientific and technological infrastructure with comparative advantage in China. Especially,the Chinese Ecosystem Research Network (CERN) leads the construction and development of the national ecosystem station network. The top-level design and overall layout have been paid more attention in the construction of the field scientific observation and research network of CAS during the 14th Five-Year Plan, and the orientation and objectives of the field station network is highlighted further. The main tasks include improving the quality of observation data continuously to enhance the capability of data services and data sharing; emphasizing the construction of key scientific infrastructure in field stations and promoting the approval of Chinese Terrestrial Ecosystem Observation and Experiment Network (Kun-Mai Project) which both are benefit to improve the equipment level and enhance the core competitiveness of the field station network; strengthening the informatization construction of the field station to enhance operational support capabilities and improve the scientific research environment; deepening the scientific research for discipline frontiers and national scientific needs to embody the value of the field station network; building and expanding the specific observation networks; and emphasizing the international exchange and cooperation.

  • Progress on Sandy Desertification Research and Future Combating Idea in Tibet, China

    Abstract: The problem of sandy desertification in Tibet is one of important regions of desertification research in China, the important progresses in the research on sandy desertification in Tibet in recent thirty years are concluded in the paper, such as finding out the status and characteristics of spatial and temporal change in recent years of sandy desertification in Tibet as well as its driving factors, establishing the theoretical system of sandy desertification in Tibet including the classification system of sandy desertification land and geological-historical evolutionary process of sandy desertification as well as the features of river valley aeolian landform in Tibet, building the combating strategy and control technical system of sandy desertification in Tibet with special plateau features. On the basic analysis of status and development trends as well as the control results of sandy desertification in Tibet, the future combating idea of sandy desertification in Tibet are forward given.

  • Construction of Long-term Ecological Research Sites in Field Station: Status, Progress and Prospect

    Abstract: Research sites are key facilities of a field station and basic conditions for conducting long-term observation, experiment, and research, which provide a great deal of direct and comprehensive data for long-term ecological research. In 2016, the Chinese Academy of Sciences started the project of construction of long-term research sites in some field stations. This project has achieved remarkable progress and provided a basis for pushing forward the construction of long-term research sites in other field stations.

  • 肝脏特异性CD36基因敲除小鼠的 制备及鉴定

    Abstract:目的 运用Cre/Loxp重组酶系统构建肝脏特异性CD36基因敲除小鼠并进行鉴定和验证,为研究CD36的生物学功能奠定基础。 方法 构建CD36打靶载体,电转转染胚胎干细胞,通过长链PCR筛选出正确同源重组的阳性克隆,阳性胚胎干细胞克隆经扩增后,注射入C57BL/6J小鼠的囊胚中,获得嵌合小鼠,再与Flp小鼠交配筛选获得Flox杂合子小鼠,该小鼠与引进的Alb-Cre小鼠交配,在F3代获得CD36fl/fl:Alb-Cre+基因型小鼠,即为肝脏特异性CD36敲除小鼠。采用PCR鉴定小鼠基因型,PCR、实时荧光定量PCR和Western blot验证小鼠肝脏CD36敲除效果,Western blot检测小鼠肾脏、脂肪和心肌组织CD36表达情况,HE染色观察小鼠肝脏形态学改变。 结果 建立了CD36基因的Flox杂合子小鼠,与Alb-Cre小鼠交配后,在F3代筛选出CD36fl/fl:Alb-Cre-和CD36fl/fl:Alb-Cre+基因型小鼠,DNA水平证实CD36fl/fl:Alb-Cre+基因型小鼠肝脏CD36基因通过Cre/Loxp重组酶系统被敲除。与CD36fl/fl:Alb-Cre-基因型小鼠相比,CD36fl/fl:Alb-Cre+基因型小鼠肝脏CD36mRNA和蛋白表达水平显著降低,肾脏、脂肪和心肌组织CD36蛋白表达无差别,肝脏形态学特征无明显差异。结论 通过Cre/Loxp重组酶系统成功构建了肝脏特异性CD36基因敲除小鼠,为研究CD36在肝脏代谢和肝脏疾病中的功能提供了动物模型
